Detection of Human Papillomavirus DNA in condyloma Acuminata using in Situ Polymerase Chain Reaction

Abstract

|
Detection of human papillomavirus(HPV) DNA in paraffin-embedded, formalin fixed tissues of condyloma acuminatum lesions by in situ polymerase chain reaction(PCR) is described. Instead of multiple primers needed for in situ PCR, one pair of
primers
were
used for amplification of HPV. In addition, direct incorporation of nonisotopically labeled dNTP was tried to eliminate hybridization step after amplification. Samples included in this study were 12 cases of condyloma acuminatum and 3 cases of
psoriasis
vulgaris as negative controls. PCR was carried out as described by Nouvo et al. with minor modifications. We withheld the addition of DNA amplification solution to the specimen until the slides were heated up to 80¡É. After 45 cycles of PCR, we
performed nonisotopic visualization without in situ hybridization procedure. Positive reaction was evident in the entire epidermis including basal layer, while positive reaction was confined to the uppermost epidermis by in situ hybridization. We
think
in situ PCR employing some modification is very sensitive for detection of HPV in condyloma acuminatum lesions.
